The crash occurred on the southbound side of the Interstate 95 near Quantico in Stafford County at about 2.35am local time as traffic slowed for a work zone, state police said.
All five of the people who died were in the vehicles hit by the bus, with three of the injured in a critical condition.
"The preliminary investigation indicates that traffic was slowing southbound for an upcoming work zone," Virginia State Police said.
"A bus failed to slow for traffic, and struck six vehicles."
It was not immediately known what the bus was being used for or how many people were on board.
The crash is under investigation and charges are pending, police said.
